A promising cancer drug, first discovered in a mushroom commonly used in Chinese medicine, could be made more effective thanks to researchers who have discovered how the drug works. The research is funded by the Biotechnology and Biological Sciences Research Council and was carried out at The University of Nottingham.

Un nouveau médicament découvert dans un champignon utilisé en médecine chinoise pourrait être rendu plus efficace grâce aux chercheurs qui ont découvert comment le médicament fonctionne.

In research to be published in the Journal of Biological Chemistry, Dr Cornelia de Moor of The University of Nottingham and her team have investigated a drug called cordycepin, which was originally extracted from a rare kind of wild mushroom called cordyceps and is now prepared from a cultivated form.

Le champignon sauvage nommé cordycept peut maintenant être cultivé.

Dr de Moor said: “Our discovery will open up the possibility of investigating the range of different cancers that could be treated with cordycepin. We have also developed a very effective method that can be used to test new, more efficient or more stable versions of the drug in the Petri dish. This is a great advantage as it will allow us to rule out any non-runners before anyone considers testing them in animals.”

Notre découverte va ouvrir la possibilité de faire des tests pour savoir quels cancers être traité avec cordycept. On a aussi développé des méthode efficaces qui peuvent êter utilisées pour tester de nouvelles formes du médicament en laboratoire.

Cordyceps is a strange parasitic mushroom that grows on caterpillars (see image). Properties attributed to cordyceps mushroom in Chinese medicine made it interesting to investigate and it has been studied for some time. In fact, the first scientific publication on cordycepin was in 1950. The problem was that although cordycepin was a promising drug, it was quickly degraded in the body. It can now be given with another drug to help combat this, but the side effects of the second drug are a limit to its potential use.

Le problème était que le médicament se dégradait rapidement dans le corps. Il peut être donné avec un autre médicament pour aider à combattre cette rapidité mais les effets secondaires de ce second médicament sont une limite à son utilisation.

Dr de Moor continued: “Because of technical obstacles and people moving on to other subjects, it’s taken a long time to figure out exactly how cordycepin works on cells. With this knowledge, it will be possible to predict what types of cancers might be sensitive and what other cancer drugs it may effectively combine with. It could also lay the groundwork for the design of new cancer drugs that work on the same principle.”
The team has observed two effects on the cells: at a low dose cordycepin inhibits the uncontrolled growth and division of the cells and at high doses it stops cells from sticking together, which also inhibits growth. Both of these effects probably have the same underlying mechanism, which is that cordycepin interferes with how cells make proteins. At low doses cordycepin interferes with the production of mRNA, the molecule that gives instructions on how to assemble a protein. And at higher doses it has a direct impact on the making of proteins.

La recherche a observé deux effets sur les cellules : À petite dose cordycepin empêche la croissance incontrôlé et la division des cellulles et à hautes doses il empêche les cellules de se coller les unes aux autres ce qui inhibe la croissance aussi. Ces deux effets proviennent probablement de la même propriété qui est qu'il interferre avec la façon dont les cellules fabriquent leurs protéines. À petites doses le cordycepine interferre avec la production de mARN et avec les molécules qui assemblent une protéine. À plus haute doses, il a in impact direct sur la fabrication de protéines.

Phellinus linteus (PL), a mushroom that has been used for many years in traditional Oriental medicine, slows the growth of highly invasive breast cancer cells by a variety of different mechanisms, according to a study published in the April 15 issue of the British Journal of Cancer.

Although new treatments and earlier detection have reduced overall breast cancer mortality, the disease remains the most deadly cancer among women ages 20 to 59. The big challenge to doctors has been in treating the most invasive breast cancers.

In their search for alternatives or additions to traditional cancer therapies such as chemotherapy and radiation, researchers have turned to natural substances with anti-cancer properties. "What I'm looking for are natural products that are able to modulate the invasiveness of cancer cells," explains Daniel Sliva, PhD, senior investigator and Director of the Cancer Research Laboratory at Methodist Research Institute in Indianapolis, Indiana. "We can go back to Mother Nature and find or rediscover something that has been here for a long time and may be effective in the treatment of cancer."

Dr. Sliva and his colleagues recently turned to their attention to PL. Past research has found that this mushroom species contains complex carbohydrates called polysaccharides, which demonstrate significant anti-cancer effects.

In the current study, Dr. Sliva and his colleagues tested the effects of PL on highly invasive human breast cancer cells in the laboratory. They found that the mushroom extract affects the growth of breast cancer cells in several ways.

First, PL interferes with the cell cycle to prevent cancer cells from proliferating. It blocks the activity of an enzyme called AKT kinase, which is responsible for the uncontrolled cell growth seen in cancer and plays a role in the formation of new blood vessels-a process called angiogenesis. "Blood vessels are important for the nutrition, and therefore the growth, of cancer," Dr. Sliva explains. "So if we can stop angiogenesis, we will block the delivery of nutrients to cancer, and this will result in the suppression of the growth and metastasis [spread] of cancer."

Dr. Sliva's team found that PL also prevented breast cancer cells from forming colonies-a marker of highly invasive cancer, as well as an indicator of its ability to metastasize. And, the mushroom inhibited three crucial events responsible for cancer metastasis: cell adhesion (the ability of the cancer cells to stick together and attach in other parts of the body), cell migration (the ability of cells to move in the body), and cell invasion (the ability of cells to penetrate through the organs in the body). "If you can inhibit one of these three factors…you can stop cancer cells from metastasizing," Dr. Sliva says.

Once they had demonstrated the effectiveness of PL against invasive breast cancer cells, Dr. Sliva and his colleagues wanted to determine whether it is safe. When they tested it in the lab, the researchers found that PL was not toxic to human cells.

LE champignon n,a pas été trouvé toxique en laboratoire mais de plus grandes études sont nécessaires pour des études cliniques.

The results of this study still need to be confirmed in animal studies and then human clinical trials. If it is found to be both safe and effective in humans, PL could be used as an adjuvant to chemotherapy and other breast cancer treatments.

Le Phellinus linteus empêcherait d’agir une enzyme nommée AKT, connue pour contribuer au développement et à la survie des cellules cancéreuses.

Si l’ancienne médecine asiatique connaissait déjà ce champignon, auquel elle prêtait le pouvoir de fortifier le corps et prolonger la vie, un groupe de scientifiques du Methodist Research Institute d’Indianapolis, aux Etats-Unis, vient tout juste de révéler le fonctionnement de son principe anti-cancer.

En focalisant leur étude sur le cancer du Le Phellinus linteus et le cordycept (champignons) 307163 , les chercheurs ont constaté des "résultats positifs" à l’utilisation de ce champignon, comme une baisse du développement incontrôlé de nouvelles cellules cancéreuses, la suppression de leur comportement agressif, et une moindre formation de vaisseaux sanguins nourrissant ces cellules.

Cependant, ces résultats restent à modérer : "nous ne sommes pas encore capables d’appliquer ces connaissances à la médecine moderne", a déclaré le docteur Daniel Sliva, un des principaux auteurs de l’étude.
Mais il ajoute : "nous espérons que notre étude encouragera d’autres chercheurs à examiner les usages des champignons médicinaux pour le traitement du cancer."
